Aging reduces the primary humoral response and the in vitro cytokine production in mice BJMBR

Aging is accompanied by a decrease in several physiological functions that make older individuals less responsive to environmental challenges. In the present study, we analyzed the immune response of female BALB/c mice (N = 6) of different ages (from 2 to 96 weeks) and identified significant age-related alterations. Immunization with hapten-protein (trinitrophenyl-bovine serum albumin) conjugates resulted in lower antibody levels in the primary and secondary responses of old mice (72 weeks old). Moreover, young mice (2, 16, and 32 weeks old) maintained specific antibodies in their sera for longer periods after primary immunization than did old mice. Antibodies specific to infectious bronchitis in broilers in Ceará state, Brazil Arq. Bras. Med. Vet. Zootec.

The experiment was carried out to determine the antibody levels to infectious bronchitis virus (IBV) in 1120 broilers of two broiler flocks, both from the same parental flock and free from previous vaccination. Forty chicks of each line were alloted to the control group and the sera were tested by indirect ELISA. The vaccination program consisted on the administration of commercial vaccines against IBV at 10 and 25 days of age. Chicks with low levels of maternal antibodies (Mab) did not show significant titers to the first vaccinal stimulus. Antibody conjugate radioimmunotherapy of superficial bladder cancer BABT

The administration of antibody conjugates for cancer therapy is now proving to be of clinical value. We are currently undertaking a programme of clinical studies using the monoclonal antibody C595 (IgG3) which reacts with the MUC1 glycoprotein antigen that is aberrantly expressed in a high proportion of bladder tumours. Radioimmunoconjugates of the C595 antibody have been produced with high radiolabelling efficiency and immunoreactivity using Tc-99m and In-111 for diagnostic imaging, and disease staging and the cytotoxic radionuclides Cu-67 and Re-188 for therapy of superficial bladder cancer. A Phase I/II therapeutic trail involving the intravesical administration of antibody directly into the bladder has now begun.

Background: A method for the selection of suitable molecular recognition element (MRE) for the quantification of human epidermal growth factor (hEGF) using surface plasmon resonance (SPR) is presented. Two types of hEGF antibody, monoclonal and polyclonal, were immobilized on the surface of chip and validated for its characteristics and performance in the quantification of hEGF. Common variable immunodeficiency (CVID) is an immunological disorder characterized by defective antibody production, recurrent infections, most notably of the respiratory tract, autoimmune phenomena and cancer. Some CVID patients may also present disturbances of the cellular immune response such as a decrease in the number and proportion of different lymphocyte populations, diminished lymphoproliferative response to mitogens and antigens, altered production of cytokines, and deficient expression of cell-surface molecules. The goal of this study was to investigate anti-Rickettsia spp. antibodies in sera of domestic chickens (Gallus gallus domesticus) of extensive breeding in Cerro Largo county, considered an endemic area for spotted fever in the State of Rio Grande do Sul, Brazil. Three hundred blood samples were collected and anti-Rickettsia spp. antibodies were evaluated by indirect immunofluorescence assay (IFA) in the sera obtained. Effect of aging and oral tolerance on dendritic cell function BJMBR

Oral tolerance can be induced in some mouse strains by gavage or spontaneous ingestion of dietary antigens. In the present study, we determined the influence of aging and oral tolerance on the secretion of co-stimulatory molecules by dendritic cells (DC), and on the ability of DC to induce proliferation and cytokine secretion by naive T cells from BALB/c and OVA transgenic (DO11.10) mice. We observed that oral tolerance could be induced in BALB/c mice (N = 5 in each group) of all ages (8, 20, 40, 60, and 80 weeks old), although a decline in specific antibody levels was observed in the sera of both tolerized and immunized mice with advancing age (40 to 80 weeks old). The present work was carried out to compare Infectious Bronchitis Virus (IBV) antibody titers in serum and egg yolk samples from laying hens. Sixty paired blood and egg samples were collected from laying hens of two farms. Serum samples were frozen, while egg yolk samples were diluted (1:500) before freezing. Serum and yolk samples were tested for the presence of IBV antibodies by indirect ELISA (commercial kit) and titers were compared by a correlation test (alpha=0.05). There was a high correlation (r=0.62) between the two kinds of samples, which means that titers of IBV antibodies in the egg yolk and in serum samples are quite the same. Epitopic Specificity of the Human Immune Response to the Invariant Region of a Polymorphic Plasmodium faciparum Merozoite Surface Antigen OAK

Merozoite surface antigen 2 (MSA2) is a Plasmodium falciparum vaccine candidate. Antibody responses to MSA2 in Melanesians naturally exposed to P. falciparum were investigated by ELISA using the recombinant MSA2 protein known as Ag 1609 and 65 overlapping synthetic peptides spanning predominantly the conserved N and C terminal regions of MSA2. Significant differences were obtained between control and malaria exposed subjects in the ability of sera to bind to Ag 1609. The median absorbance obtained with control sera was 0.36 whereas that obtained with sera from malaria exposed subjects was 1.8. The sera of ninety-five percent of malaria exposed subjects reacted significantly with Ag 1609. The percentage reactivity of the control sera with Ag 1609 was 39%....

Vitellogenin (VTG) was isolated from the haemolymph of Penaeus vannamei by a three step procedure including ultracentrifugation, gel filtration and ion exchange chromatography. VTG was used to raise polyclonal antibodies that were purified by ion exchange chromatography. A two step competitive assay was developed in which VTG could be quantitated by its capacity to inhibit the binding of antibody to the VTG previously adsorbed onto a solid phase. Binary mixtures of 11-mercaptoundecanoic acid (MUA) and other thiols of various lengths and terminal functions were chemisorbed on gold-coated surfaces via S-Au bonds to form mixed self-assembled monolayers (SAMs). Several values of the inole fraction of MUA in the thiol mixtures were tested and the structure and composition of the resulted thin films were characterized by X-ray photoelectron spectroscopy (XPS) and polarization modulation infrared reflection-absorption spectroscopy (PM-IRRAS). Plants from the genus Alternanthera are thought to possess antimicrobial and antiviral properties. In Brazilian folk medicine, the aqueous extract of A. tenella Colla is used for its anti-inflammatory activity. Abstract The aim of this study was to evaluate the impact and mechanism of co-blocking of costimulatory signals CD28-B7-CD40-CD40L during immune allograft rejection. Forty-eight recipient rabbits were prepared as a high-risk corneal allograft model. After surgery, the animals were randomly divided into: control group, MR1 group, anti-B7 group, and co-blocking group (n=12, each group). In this study, we compared the levels of neutralizing antibodies induced by inactivated rabies vaccine in cattle by using three alternative immunization procedures. Forty-five bovines (breed nelore) were then organized in three groups (A, B and C, with 15 animals/group). Group A received only one vaccine dose at day zero and Group B received the first dose at day zero and then another dose at day 30 (early booster). The aim of this study was to determine what the difference in performance is between positive (progeny of breeders vaccinated or with natural infection) and negative broilers to the presence of antibodies against the CAV in currently intensive raising systems.
